Resident Evil 6 là chuỗi những sự kiện về các cuộc tấn công khủng bố bằng vũ khí sinh học - BOW vào Châu Âu, Mỹ và Trung Quốc mà người đứng đằng sau Ada Wong. Mang trong mình những lý do công tư lẫn lộn, cả Chris và Leon đều lần theo dấu vết của cô để tìm ra câu trả lời cuối cùng cũng như ngăn chặn sự tàn phá của BOW.
Với campaign dàn trải ra 4 phần, cốt truyện của Resident Evil 6 là một chuỗi những những sự kiện đan chéo vào nhau khiến người chơi buộc phải chơi hết hoàn toàn để hiểu được. Cách dẫn truyện theo kiểu một sự kiện được nhìn nhận ở hai góc độ của hai nhân vật tại một thời điểm hoặc những biến cố do nhân vật trước tạo ra để lại câu hỏi hay cậu trả lời cho nhân vật đang theo đuôi thật sự rất hay.
Về phần thiết kế nhân vật, thật khó mà chê trách được Capcom khi mà những con quái trong Resident Evil 6 tỏ ra gớm ghiếc, chi tiết và ấn tượng hơn hẳn so với những mà họ đã thể hiện ở Resident Evil 5. Boss khổng lồ với những lần biến hình ghê gớm hơn, tinh xảo hơn phức tạp hơn. Môi trường game với khung cảnh thành phố ngập trong biến lửa, những vụ nổ lớn thật sự đã nâng tầm Resident Evil lên một đỉnh cao mới. Đã lâu lắm rồi, Capcom mới lại mang đến những đoạn Cutscene mang đậm tính thảm họa của đại dịch zombie như thời Resident Evil 2, Resident Evil 3
Chế độ chơi Co-op online và offline vẫn có mặt trong game lần này. Khá nhiều trường đoạn sẽ rất hay khi bạn chơi chung cùng người khác hơn là AI, như việc trèo leo giữa những tòa nhà, một người sẽ phải bắn trả để ngăn chặn bước tiến của Boss đang cố leo lên bắt lấy người chơi còn lại. Hay bạn đang cố gắng gài mìn và cần sự bảo vệ trên không của đồng đội khi các BWO đang cố gắng tiến gần. Hầu hết những trường đoạn dạng này sẽ rất dễ dàng đến độ bạn không cảm nhận được khó khăn của nó vì AI tỏ ra làm công việc mình rất tốt. Nhưng nếu như đó là sự phối hợp của người và người thì đó lại là chuyện khác.
